Abstract: Microdroplet has become an interesting media for acceleration of various reactions. Recently, in collaboration with both Agilent Team and Dr. Harsha Gunawardena from J&J, we reported an ultrafast microdroplet-based digestion approach for antibody including mutated antibodies and ADC drugs in which the digestion can complete within 1 ms. Such fast digestion thus facilitates rapid and high throughput characterization of antibody protein therapeutics by high resolution mass spectrometry. On the other hand, in collaboration with Dr. Shawn Wu at Merck Co., antibody proteins including impurity host-cell proteins (HCPs) can be also quantified by coulometric mass spectrometry (CMS), a combined electrochemistry and mass spectrometry technique (EC/MS), which can be used to perform absolute quantitation of small organic molecules, peptides and proteins as well as protein post-translational modifications, without the use of standards.
Bio: Hao Chen graduated from Wuhan University in 1996 and obtained his Ph.D. degree from Purdue University in 2005. He is currently a professor in chemistry at New Jersey Institute of Technology (NJIT). Dr. Chen performs research on organic and biological mass spectrometry for organic reaction mechanism elucidation, bioanalysis and instrument development and has published 160+ papers in peer-reviewed journals (H index 52). Recently his lab has developed several rapid methods for detecting and degrading PFAS and microplastics/nanoplastics. His lab also developed a coulometric mass spectrometry (CMS) method for absolute quantitation of peptides, proteins and organic compounds, without the need of using standards or calibration curves.
